Electrochemical immunosensor for detection of CA 15-3 biomarker in point-of-care

This work reports the development of a simple and rapid electrochemical immunosensor for determination breast cancer biomarker Cancer Antigen 15–3 (CA 15–3). Disposable cost-effective chips, consisting gold screen-printed electrodes (AuSPEs), were used to develop portable devices monitoring in point-of-care (PoC), under clinical context. The biosensor preparation consisted two steps. First, self-assembled monolayer (SAM) mercaptosuccinic acid (MSA) was formed at AuSPE surface. Then, CA antibody covalently bound carboxylic groups standing electrode surface using EDC/NHS chemistry. performance developed evaluated by assessing sensor sensitivity, linear response interval, selectivity detection limit (LOD). provided wide concentration range (from 1.0 1000 U mL−1) low levels achieved (LOD 0.95 mL−1), enabling sensitive clinically relevant levels, square wave voltammetry (SWV) as electroanalytical technique. Moreover, studies performed against other biomarkers 125 19–9) revealed that has high antigen. applied quantification artificial serum samples with satisfactory results.
